The link reaction in aerobic respiration is the formation of acetyl CoA which is used in Krebs cycle. The oxidative decarboxylation of pyruvate into acetyl CoA involves the presence of atleast five essential cofactors and a complex enzyme. The cofactors involved are Mg ions, thiamine pyrophosphate (TPP), NAD⁺, coenzyme A and lipoic acid. Thiamine (Vitamin B₁) was isolated from rice polishing and is known to occur in active form (thiamine pyrophosphate) or as coenzyme of certain enzymes.
